چکیده
Technical aspects of rare-event detection are discussed in this article in a practical context, with two real-life examples. A growing number of flow cytometry-based assays depend on rare-event detection for basic science and clinical applications.

On the possibility of automated scoring of pollen mutants.
Both flow cytometry and automated image analysis techniques may be useful in relieving the drudgery, increasing the speed, and adding quantitation to the scoring of pollen mutants, but practical problems related to the detection of very rare events have to be overcome. The features of flow and image cytometry instrumentation that may be useful for pollen measurements are discussed and a qualita...

Rare-event analysis (REA) is the art of finding a needle in a haystack, confirming that it is a needle, and making measurements to determine what kind of needle it is. A rare event is typically one occurring at a frequency of below 1 in 1,000 (0.1%) [1, 2]. Numerous measurements are needed to detect rare events, such as tumour cells in peripheral blood, and it was historically a long-winded pro...
متن کاملHierarchical Modeling for Rare Event Detection and Cell Subset Alignment across Flow Cytometry Samples
Flow cytometry is the prototypical assay for multi-parameter single cell analysis, and is essential in vaccine and biomarker research for the enumeration of antigen-specific lymphocytes that are often found in extremely low frequencies (0.1% or less). Standard analysis of flow cytometry data relies on visual identification of cell subsets by experts, a process that is subjective and often diffi...

متن کاملOptimization of a flow cytometry-based protocol for detection and phenotypic characterization of multipotent mesenchymal stromal cells from human bone marrow.
BACKGROUND To study the biology of rare bone marrow (BM) multipotent mesenchymal stromal cells (MSCs), recognized protocols are needed. Colony-forming unit-fibroblast (CFU-F) assays have historically been used for the enumeration of MSCs. However, the need to isolate and further analyze MSCs requires new strategies based on cell surface markers. The purpose of this work was to verify the phenot...
